Penapisan Dan Uji Antagonis Jamur Endofit Asal Tanaman Akasia (Acacia Crassicarpa) Terhadap Ceratocystis Sp. Secara In-Vitro Serta Identifikasinya
ABSTRACT
This study aims to select the endophytic fungi of acacia plants which have
the best antagonistic potential as biological agents against fungi Ceratocystis sp..
The study was conducted at the Laboratory of Plant Protection Departement
PT. Arara Abadi, Desa Pinang Sebatang, Kecamatan Tualang, Kabupaten Siak,
Riau. The study was conducted for six months from March 2019 to August 2019.
This study was conducted using exploration, observation and experiment methods
with in vitro tests consisting of 6 stages, namely: isolation and purification of
endophytic fungi from acacia plants, antagonist test of endophytic fungi from
acacia plants against Ceratocystis sp., hypovirulence test, growth test. endophytic
fungi from acacia plants which are antagonistic against Ceratocystis sp.,
hyperparasitism test and identification of endophytic fungi from acacia plants
which are antagonistic against Ceratocystis sp.. The data obtained from
experiments 1, 5 and 7 were analyzed descriptively and presented in the form of
tables and figures. Experiments 2, 3 and 4 were analyzed by analysis of variance
and to compare the mean between isolates followed by Ducan's new multiple
range test (DNMRT) at the 5% level. The results of the isolation of the acacia
plant endophytic fungi obtained 12 isolates. Total of 6 isolates of endophytic
fungi with high antagonistic power, 5 isolates were hypovirulent fungi and
1 isolate was virulent. E8 isolate had the highest antagonist power, namely
54.37%. E2 isolate had the highest growth rate of antagonistic fungi (31.28
mm/day). Endophytic fungal hyperparasitic type with Ceratocystis sp. namely
attachment of isolate E8, entanglement of isolate E7, entanglement of isolate E4,
isolate isolate E1 and E2. The identification results showed that isolates E1, E2 and
E4 belonged to the genus Trichoderma, isolates E7 and E8 belonged to the genus
Fusarium.
