Pengaruh Suhu Berbeda Terhadap Aktivitas Ekstrak Kasar Enzim Kolagenase Organ Dalam Ikan Nila Salin (Oreochromis Niloticus)
ABSTRACT
Enzymes are biocatalysts that function to accelareta the rate of a chemical reaction without being involved in the reaction. Collagenase is a proteolytic enzyme capable of degrading polypeptide bonds. The activity of the enzyme hydrolyzing protein is influenced by several factors, namely temperature, pH, substrate, inhibitor, and activator. This study was aimed to determine the effect of temperature differences on the activity of the crude extract of the collagenase enzyme from the internal organs of saline tilapia (Oreochromis niloticus). The research method esed was an experiment with a completely randomized design (CRD) non-factorial. The treatments used were different temperatures of 45ºC, 55ºC, and 65ºC. The parameters of the analysis were yield, concentration of dissolved protein, and the activity of crude extract of collagenase enzyme from the internal organs of saline tilapia (Oreochromis niloticus). The results showed that the internal organs of saline tilapia (Oreochromis niloticus) had a yield value of 40.54% and a soluble protein concentration of 19.39 mg/mL. Activity of enzyme collagenase from crude extract of internal organ saline tilapia at 45ºC, 55ºC, and 65ºC 0.297 U/mL, and 0.272 U/mL, respectively. The highest crude extract activity of collagenase enzyme was 0.326 U/ml at 55ºC,
