EXPRESSION OF MONOCYTE CHEMOATTRACTANT PROTEIN-1 (MCP-1) ATHEROSCLEROTIC RATS AORTA AFTER ADMINISTRATION OF EXTRACT OF MANGROVE FRUIT (Sonneratia alba)

Sonneratia alba is a plant that is high in antioxidants such as alkaloids, phenolics, flavonoids, tannins, steroids, terpenoids and saponins. Antioxidants has been proven can prevent foam cell formation, in which MCP-1 plays role in the foam cell formation by recruiting monocytes that take in oxidized-LDL. This study aimed to analyze the effect of mangrove Sonneratia alba fruit extract on expression of MCP-1 in early stages of atherosclerosis in rats fed a high-fat diet. Sixteen paraffin blocks of Rattus norvegicus rats’s abdominal aorta were used in this study which consisted of 3 groups: standard diet (KS), high-fat diet (KP), and high-fat diet and Sonneratia alba fruit extract (KT). Sonneratia alba fruit extract effect on expression of MCP-1 in abdominal aorta was assessed by immunohistochemistry examination. Expression of MCP-1 area was counted in 5 fields of microscopic view and evaluated using the Photoshop Image Analysis program. The results showed that the average percentage of MCP-1 expression in the KS, KP and KT groups was 6,32 ± 1,27%, 24,59 ± 9,68% and 17,45 ± 6,93%. Analysis using One Way Anova showed differences in the area of expression in the three groups, but not significant (p>0.05). Sonneratia alba mangrove extract was not able to significantly suppress MCP-1 expression in the early stages of atherosclerosis.
