ABSTRACT
Rotan getah is a species of plant from the Arecaceae family which belongs to the large-sized rattan group with a height of up to 40 m. The DNA barcoding analysis using the trnL-trnL-trnF intergenic spacer sequence in this plant has never been done. The advantage of this sequence’s that it has a high level of variation due to the high number of mutations. This research aims to analyze the sequence of DNA barcoding in the trnL-trnL-trnF IGS in rotan getah from Kampar. Total DNA extraction by using Genomic DNA Mini Kit Plant (Geneaid). The stages of the research are total DNA isolation, PCR (Polymerase Chain Reaction), total DNA and amplicon electrophoresis, sequencing, and data analysis. The morphological observations showed that the morphology of this rotan getah was similar to that of C. angustifolius Griff. The results of the analysis showed that the trnL-trnL-trnF IGS sequence obtained from the three samples each had a size of 867 bp. Rotan getah has the highest genetic similarity (100%) with Calamus sp. 'Kajuik Lake' and lowest with Oncocalamus mannii. The results of this study also confirm that rotan getah and Calamus sp. 'Kajuik Lake' is the same species with scientific name C. angustifolia Griff.
