Pengaruh Naa, Bap Serta Kombinasi Naa Dan Bap Pada Media Ms Terhadap Pertumbuhan Eksplan Daun Sansevieria Robusta Secara In-Vitro
ABSTRACT
Sansevieria or tongue-in-law from the Agavaceae family is an ornamental plant that can be used as fiber material, medicinal plants and can absorb air pollution or harmful chemical compounds such as formaldehyde, benzene and trichloroethylene. The many benefits contained in Sansevieria cause the demand for this plant to be high, but the growth period of this plant is relatively slow so that propagation methods such as tissue culture are needed in order to produce Sansevieria plants quickly in large quantities and uniformly. The purpose of this study was to see the effect and get the best treatment concentration of NAA, BAP and the combination of NAA and BAP for the growth of Sansevieria robusta leaf explants in-vitro. This study was conducted from January to May 2022 using a completely randomized design consisting of 8 treatments and 3 replicates so that there were 24 experimental units. The parameters observed were when callus appeared, callus color, callus texture, callus weight, percentage of explants producing callus, when shoots appeared, shoot length, number of shoots, percentage of explants producing shoots. Data were analyzed using variance analysis and further tested using Duncan's multiple range test at the 5% level. The results showed that the concentration of NAA, BAP and the combination of NAA and BAP on MS media influenced the growth of Sansevieria robusta leaf explants, namely when callus appears, callus weight, percentage of explants producing callus, when buds appear, bud length and percentage of explants producing buds. The treatment with NAA 2 mg.l-1 + BAP 2 mg.l-1 is the best treatment for the growth of Sansevieria robusta leaf explants when callus appears, callus weight, percentage of explants producing callus, when shoots appear, shoot length and percentage of explants producing shoots.
