Uji Kemampuan Isolat Aktinomisetes Lokal Asal Siak Dan Indragiri Hilir Dalam Menghasilkan Enzim Ligninase, Selulase Dan Amilase

ABSTRACT
Enzymes are one of the natural products that play an important role in various
industrial applications, like a food processing, chemical product processing, textile
industry, pulp and paper industry, medicine field and recombinant DNA
technology. Seeing the importance of several enzymes such as ligninase, cellulase,
and amylase in various fields, there have been many previous studies that have
tested the ability of actinomycetes in producing these enzymes. This study aims to
test and select the ability of 22 actinomycetes isolates (16 isolates from the
rhizosphere of the Siak-siak plant (Dianella ensifolia L.) and six isolates from
mangrove soil in Kuala Enok, Tanah Merah Regency, Indragiri Hilir) in producing
ligninase, cellulase, and amylase enzymes by qualitative assay was indicate by the
formation of a clear zone around the colony and descriptive assay by count
enzymatic index of isolates. Assay the ability of actinomycetes isolates in
producing ligninase, cellulase, and amylase enzymes using dotted method on the
selective medium of each type of enzyme with different substrate. Based on the
results obtained, 16 isolates are capable of producing ligninase with the highest
enzymatic indes of 2.51 owned by A2, 13 isolates are capable of producing the
enzyme cellulase with the highest enzymatic index of 2.02 owned by RW8, and 14
isolates are capable of producing the enzyme amylase with the highest enzymatic
index of 4.42 owned by RK4.
