Analisis Sekuens Nucleotide Binding Site-Leucine Rich Repeat (Nbs-Lrr) Kopi Liberika (Coffea Liberica W. Bull Ex. Heirn) Dari Kepulauan Meranti
ABSTRACT
Coffee Leaf Rust (CLR) disease is one of the main diseases of coffee plants including Liberica coffee. Liberica coffee is superior variety from Meranti Islands, Lim 1 and Lim 2, have been determined by Minister of Agriculture as resistant varieties against Coffee Leaf Rust (CLR). The largest group of resistance (R) genes is the genes encoding the NBS-LRR (Nucleotide Binding Site-Leucine Rich Repeat) protein. The purpose of this study was to analyze the NBS-LRR region of Lim 1 and Lim 2. In this study, PCR products using degenerate and non-degenerate primers were sequenced and subsequently converted into amino acid sequence. Results of this study indicate that deduced amino acid sequences contain conserved domain of NB-ARC after being converted using the Conserved Domain Database (CDD). In addition, there are three motifs i.e., Kinase-2, RNBS-B and GLPL. The sequences obtained in this study have the potential as genetic markers for resistance in coffee.
