ANALISIS SEKUENS BARKODE DNA PADA DAERAH Matk TUMBUHAN LOPANG (Gymnopetalum Cochinchinense L.)
ABSTRACT
Lopang is an annual herbaceous plants that have been cultivated in Riau. This plant is also known by several local names, such as lopang, kanjat, and wild bitter melon. The scientific name of Lopang is Gymnopetalum cochinchinense L. Kurz. The scientific name for this species is still unclear due to the use of various common names and limited literature. This study aims to analyze the DNA sequences of matK region in lopang plants. The stages of this research included morphological observation, sample preparation, total DNA isolation using Genomic DNA Mini Kit Plant (Geneaid), Polymerase Chain Reaction (PCR) using matK-413f-1 and matK-1227r-3 primer pairs, electrophoresis of PCR products, sequencing, and data analysis using BioEdit, BLASTn, and MEGA version 6. Morphological observations of lopang indicated that lopang was a monoecious herb with a taproot system that grows vines. Lopang has tendrils, palmate-shaped leaves, white petals, ellipsoid with reddish-yellow to red coloured fruit, and numerous seeds. The results showed that the lopang sequence was 752 bp in length. The results of the BLASTn analysis showed that Gymnopetalum chinense had the highest similarity (99.73%) with individual lopang plants studied. The dendrogram showed that the lopang studied forms a group that was separated from the genus Trichosanthes. The matK DNA sequence of lopang plants obtained in this study was the first to be reported and can be used as a reference for identifying lopang.
