Induksi Perakaran Pada Tunas Dendrobium Black Spider X Dendrobium Violaceoflavens ‘Paras Ayu’ Dengan Penambahan Jenis Dan Konsentrasi Auksin Yang Berbeda Secara In Vitro
ABSTRACT
Dendrobium Black Spider x Dendrobium violaceoflavens 'Paras Ayu' is a type of hybrid orchid found in Indonesia. This orchid was registered with the RHS (Royal Horticultural Society) under the trade name Dendrobium Paras Ayu in 2018. Root induction was carried out by thinning shoots on new media by adding a growth regulator from the auxin group in vitro. The purpose of this study was to determine the effect of auxin types, concentrations and interactions between the two on the in vitro induction of D. Paras Ayu orchids. This study used the completely randomized factorial design with two research factors. First, the type of auxin with 3 treatment levels (K1 = IBA, K2 = NAA, and K3 = 2.4-D) and other factor was the concentration of auxin with 4 treatment levels (I0 = control, I1 = 0.25 mg/L, I2 = 0.5 mg/L, and I3 = 0.75 mg/L). There are twelve treatment combinations with five repetations, and sixty experimental units. Moreover, there were four parameters of observation, such as the precentage of live explants, time of root emergence, number of roots, and root length. Data were analyzed by Analysis of Variance (ANOVA) and continued with Duncan's Multiple Range Test (DMRT) at a 5% level. The results of this research indicated that the interaction between the type and concentration of auxin have the significant effect on the percentage of live explants and root length of D. Paras Ayu orchids. The IBA treatment with the concentration of 1.25 mg/L showed the best farming roots with 100% live explant percentage, with the highest average number of roots (0.80 pieces) and the longest roots (0.50 cm) on Murashige Skoog media.
