Analisis Sekuens Leucine-Rich Repeat Receptor-Like Kinase (Lrr-Rlk) Kopi Liberika (Coffea Liberica W. Bull Ex. Hiern) Dari Kepulauan Meranti
ABSTRACT
Liberica coffee (Coffea liberica W. Bull Ex. Hiern) is one of the plants that have been developed in peatlands area such as Riau. One of factors that reduce the productivity of coffee plants is pests and plant diseases. Liberica coffee varieties Liberoid Meranti 1 (Lim 1) and Liberoid Meranti 2 (Lim 2) are from Kepulauan Meranti and resistant to leaf rust disease. LRR-RLK sequence in Liberica Meranti coffee has not been analyzed. This study aimed to analyze the LRR-RLK sequences of Lim 1 and Lim 2 Liberica coffee. Total genomic DNA was amplified using LRR_RLK2 primer pair. Subsequently PCR product was sequenced and analysed. Result showed that the primer could amplify and produced a 400 bp bands. In addition, deduced amino acid sequences contain PLN00113 conserved domain and LRR conserved motif. These results indicate that the LRR-RLK marker can be used to detect leaf rust disease resistance in Liberica coffee plants.
