Analisis Barkode Dna Dan Morfometrik Tikus Dari Gunung Mekongga Sulawesi Tenggara
ABSTRACT
The high diversity of Muridae in Mekongga Mountain, Southeast Sulawesi, requires identification methods. Identification methods can be through morphometric and molecular approaches. This study aims to analyze the type of mice in Mount Mekongga, Southeast Sulawesi using molecular approach of DNA barcode of CO1 gene and measurement of 20 morphometric characters of mouse skull. The research process includes the measurement of rat skulls, Principal Component Analysis (PCA) with the PAST software program, total DNA isolation, PCR that amplifies the CO1 gene fragment and sequencing. The results of the study three main components (KU1, KU2, KU3) contributed 88.81% of the variance. Morphometric characters that act as the main differentiators are GLS (largest skull length), ZB (zygomatic width) and LN/LON (nose length). DNA barcoding sequences of the CO1 gene were able to group 16 mouse species from the same 8 genera into members of the Muridae family.
