Isolasi Dan Uji Aktivitas Antibakteri Senyawa Metabolit Sekunder Ekstrak Etil Asetat Tumbuhan Paku Tanduk Rusa (Platycerium Bifurcatum)
SUMMARY Platycerium bifurcatum is an epiphytic fern, which belongs to the family Polypdiceae and genus platycerium. Other species of the genus Platycerium, such as P. angolense, P. coronarium, P. grande and P. superbum have previously been reported for their potential bioactivity as antioxidants, antibacterials, anti-inflammatory and anticancer. In this species, there is not much information about bioactivity tests. This study aims to isolate secondary metabolite compounds against ethyl acetate extracts of plants P. bifurcatum and know the potential of the compound as an antibacterial substance. Samples were extracted by maceration method using methanol solvent and fractionated by liquid-liquid extraction using n-hexane solvent, dichloromethane, ethyl acetate. The ethyl acetate fraction was separated by VLC to produce ten main fractions, in fraction IV a compound was obtained coded 22-09-C-FIV. Fractions 7,8 and 9 were combined and separated using gel chromatography to produce six main subfractions. In subfraction VI a compound is obtained, coded 22-09-C-FVII-SF6-64/108. The purity of the compound was tested by thin layer chromatography (TLC), Fisher Johns melting point test and high performance liquid chromatography (HPLC) analysis. The compound 22-09-C-FIV was characterized and elucidated using UV-Vis spectroscopy, FTIR and NMR obtained as kaempferol based on comparative data. The compound 22-09-C-FVII-SF6-64/108 was characterized using UV-VIS, FTIR and NMR spectroscopy, the characterization results showed that the compound was ferulic acid. Both compounds were tested for antibacterial activity using the resulting microdilution method that has good potential in inhibiting growth of pathogenic bacteria.
