Analisis Hubungan Ekspresi Gen MSH3 Dengan Prognosis Pasien Karsinoma Endometrial
ABSTRACT
Endometrial carcinoma is the most common malignancy in the deepest part of the uterus (endometrial). Failure of the DNA repair systems such as Mismatch repair (MMR) will occur microsatellite instability (MSI) if it do not repaired properly and is associated with endometrial carcinoma. MSH3 is one of the MMR genes which plays a role in the DNA repair system. The aim of this study is to analyze association between gene expression and the prognosis of endometrial carcinoma patients. This research is an observational analytical study with a retrospective cohort design. Clinical profiles data was taken from The Cancer Genome Atlas (TCGA) database, MSH3 gene expression data was taken from GDC TCGA-UCEC and molecular classification data from cBioPortal using total sampling technique. This research used 529 samples and will be analyzed using univariate, bivariate (Kaplan Meier) and multivariate (Cox regression) methods. Based on statistical tests, there is a significant association between MSH3 gene expression with grading, histopathological type and molecular classification of endometrial carcinoma patients. This study also found a significant unfavorable relationship between overall survival of endometrial carcinoma patients and MHS3 gene expression, but not significant on progression free survival. Stage and grading are independent factors for the prognosis of endometrial carcinoma patients. In conclusion, high MSH3 gene expression is associated with poor prognosis of endometrial carcinoma patients and MSH3 gene expression is not an independent factor for the prognosis of endometrial carcinoma patients
