Respon Pertumbuhan Eksplan Petiol Tembesu (Fagraea Fragrans Roxb.) Dengan Penambahan Thidiazuron (Tdz) Secara In Vitro
Tembesu (Fagraea fragrans Roxb.) is a potential wood plant that is commonly used in heavy and domestic construction. The bark of the stem, branches and leaves of tembesu can also be used as a medicine to reduce heat and dysentery. The demand for tembesu that increases from year to year without being followed by tembesu cultivation results in a decrease in tembesu stands in nature. Planting tembesu using natural seed is not enough because of the limited amount. One of the multiplications that can be done is through culture in vitro. This study aims to determine the effect of giving TDZ on the growth of tembesu petiole explants and determine the best TDZ concentration. This research was carried out at the Plant Tissue Culture Laboratory, Biology Department, Faculty of Mathematics and Natural Sciences, Riau University from July-August 2023. The design used was a Completely Randomized Design (RAL) consisting of 5 treatment levels (0, 0.5, 1 , 1.5 and 2 mg/L TDZ) with 5 repetitions. The results showed that the addition of TDZ responded to the percentage of live explants. The highest percentage of live explants was 90% in treatment P1 (0.5 mg/L TDZ).
Key finding: Fagraea fragrans, in vitro, petiole, TDZ
