Respons Pertumbuhan Eksplan Nodus Kotiledon dari Nucellar Seedling Jeruk Siam (Citrus nobilis Lour.) dengan Penambahan NAA dan BAP pada Media MS
This research aims to determine the interaction effect of NAA and BAP, NAA factor, and BAP factor, and to obtain the best treatments on the growth of Siamese orange cotyledonary node explants on MS media. This research was conducted experimentally using factorial randomized block design with two factors: NAA (0; 0,25; 0,50; and 0,75 mg.l-1) and BAP (1, 2, 3, and 4 mg.l-1). Parameters observed were shoot emergence, number of shoots, shoot height, number of leaves, and percentage of explant survival. The data were analyzed using analysis of variance followed by Duncan's New Multiple Range Test (DNMRT) at 5% level. The results showed that the interaction of NAA and BAP increased the number of leaves with the best treatment was 0,25 mg.l-1 NAA and 1 mg.l-1 BAP produced 6,33 leaves. Application of NAA had no significant effect on all observation parameters. Application of 3 mg.l-1 BAP gave the best results at the time of shoot emergence (5,58 days after inoculation) and the number of shoots (4,26 shoots). The percentage of explant survival ranged from 88,89 – 100% and consisted of three types of regeneration, which were shoots only, callus only, and both shoots and callus. This research also found somatic embryogenesis that occurred in callus with compact texture and greenish color at the treatment of 0 mg.l-1 NAA and 3 mg.l-1 BAP.
