Analisis Sekuens Barkode Dna Matk Dan Trnl-Trnl-Trnf Intergenic Spacer (Igs) Pada Tumbuhan Rotan Pahit (Calamus Caesius L.) Asal Riau
Bitter Rattan Plant (Calamus caesius L.) is one of Arecaceae family types that has a shrub habitus characterization, growing in clumps, and creeping on other plants. This plant is usually taken by the Rokan Hilir citizens as a traditional culinary called 'pangkek'. The aim of the study is to analyze the sequences barcodes DNA of matK and trnL-trnL-trnF IGS of bitter rattan plant (Calamus caesius L.). The methods of the research were total DNA isolation, electrophoresis, PCR (Polymerase Chain Reaction), PCR product electrophoresis, purification and sequencing, and data analysis of using BioEdit version 7.0, BLASTn, and MEGA 6.0. The results of this research showed that DNA barcode matK of bitter rattan (Calamus caesius L.) is 862 pb. It has the highest similarity to Calamus walkeri (99.07%). the sequences DNA barcode of trnL-trnL-trnF IGS is 910 pb. It is closely to the sequences of Calamus walkeri (99.23%). In this research, the identity of value was not able to be reached to100% yet. The result has showed that the sequences genes of DNA matK and trnL-trnL-trnF IGS of bitter rattan plant from Rokan Hilir is unavailable yet in the GeneBank database.
