People tend to have a less productive lifestyle, excessive use of electronics and consumptive behavior that affects sleep patterns. Bad habits carried out continuously cause hormonal imbalances characterized by insomnia and depression. The amino acid L-Tryptophan (Trp) is reported to improve sleep quality and early treatment of early symptoms of insomnia without depressant drugs. However, the use of Trp should be in appropriate doses as the effects of overdose are quite serious. Therefore, monitoring of dosage levels on supplement packaging containing Trp is necessary to minimize adverse reactions. This study used an amperometric electrochemical biosensor method combined with the metabolism of Saccharomuces cerevisiae yeast. The sensor was designed using Pt-Ag electrodes with data results in the form of voltage proportional to dissolved oxygen (DO). Trp samples consisted of a variety of concentrations including 100 mM, 200 mM, 300 mM, 400 mM and 600 mM and supplements containing Dr. Berg SLEEP AID commercial Trp. The resulting DO levels for the various Trp concentrations were 41.76%; 34.26%; 30.65%; 25.04% and 16.36%. Furthermore, the measurement of Dr. Berg SLEEP AID supplement resulted in a DO level of 37.7%. The results show that the yeast-based amperometric biosensor can detect changes in Trp concentration as it affects the activity of yeast consuming oxygen. Further data analysis using PCA resulted in a visualization score plot with a total variance of 87.58% with 68.93% for PC1 and PC2 values of 18.65%. This reflects that biosensor data can be combined with PCA.
