Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Ekstrak Etil Asetat Daun Taxus Sumatrana
Taxus sumatrana belongs to the Taxaceae family and the Gymnospermae sub- division. In Indonesia, this plant is known as the cemara Sumatera, tampinur batu and kayu taji. This plant has been known as a plant that contains anticancer and antioxidant compounds. This study aims to isolate and characterize secondary metabolite compounds from ethyl acetate extract of T. sumatrana leaves and test toxicity using the brine shrimp lethality test (BSLT) method. Isolation of this plant was carried out using VLC (vacuum liquid chromatography) and produced 12 fractions. Fractions 1-7 were combined and further fractionation was carried out using flash column chromatography and 17 main subfractions were produced. The results of flash column chromatography fractionation in subfraction 5 were purified and yellow crystals were obtained in subfraction 5 weighing 8 mg and coded TS- SF5. The purity of the compound was tested using thin layer chromatography (TLC) and produced a single spot and a melting point test was obtained around 290-292oC. Purity analysis was also carried out using a high performance liquid chromatography (HPLC) instrument which showed a dominant peak at 22.076 minutes. The TS-SF5 compound was subjected to UV-Vis and FTIR spectroscopy analysis. The UV-Vis spectrum of the TS-SF5 compound showed absorption bands at wavelengths of 329, 271 and 218 with absorbance values of 0.87, 1.01 and 1.31. The results of the IR spectrum of the TS-SF5 compound showed the presence of several functional groups at wave numbers 3135 cm-1 (OH), 3075 cm-1 (aromatic C-H) 2845 cm-1 (aliphatic C-H) and 1649 cm-1 (aromatic C = C). The results of the toxicity test of the TS-SF5 compound obtained an LC50 value of 5.309 ppm. This shows that the TS-SF5 compound is very toxic.
