Optimasi Produksi Lakase Dari Trichoderma Asperellum Lbkurcc1 Dengan Penambahan Ion Cu2+ Pada Fermentasi Padat Dengan Substrat Serbuk Gergaji Kayu Keruing
Laccase is environmentally friendly extracellular enzyme and is commonly called a multicopper enzyme because it contains 4 copper ions on its active side. Laccase has a broad spectrum so that it can be applied in various industrial fields. Laccase can be produced from mushrooms, one of which is the fungus Trichoderma asperellum LBKURCC1. The purpose of this study is to produce lacase enzymes and determine the activity of lacase by using ABTS as a substrate and determine the protein content to obtain specific activities. Laccase production is carried out by means of solid fermentation using keruing sawdust as a substrate by adding variations in the concentration of Cu2+ ions (0; 0.5; 1.0; 1.5 and 3 mM). Fementation was carried out for 8 days and then extracted with 0.05 M Na-acetate buffer pH 5.5 and lacase activity test was performed using ABTS as a substrate. The crude enzyme extract obtained was then concentrated and washed with a 0.05 M Na-acetate buffer of pH 5.5 and a laccase activity test was performed. Determination of specific activity is carried out by testing the protein content of each enzyme by using the Lowry method. The results showed that the highest lacase activity in addition of Cu2+ 1.5 mM ions was 0.4361 U/L in crude enzyme extracts and 3.3902 U/L in 5x concentrated enzymes, however, the addition of Cu2+ ions did not provide distinct activity. significant (P
