Isolasi, Uji Aktivitas Antioksidan Dan Toksisitas Senyawa Metabolit Sekunder Dari Tanaman Miana Merah (Coleus Hybridus)
Red miana (Coleus hybridus) is one of species in genus Coleus, family Lamiaceae which is a potential source of traditional medicines. The purpose of this study is to isolate, examine biological activities, including antioxidant and toxicity from secondary metabolites of the plant. As much as 13.7 kg C. hybridus was extracted with maceration method by using methanol and separated into two fractions by n-hexane and ethyl acetate. The purification of compounds was conducted by flash chromatography. This process has afforded two crude compounds. First compound was observed as green crystalline compound, obtained from fractions 9 to 15 and it was designated as CH-5. Second compound was obtained from fractions 96 to 101. Based on the comparison of spectral data, CH-5 was determined as ferulic acid and CH-6 was 12α-7α,18β-trihidoksi-13β,16-cyclo-8-abietene-11,14-dion-12-O-β-D-glucoside. Extract n-hexane, extract ethyl acetate, CH-5 and CH-6 was analysed for their radical scavenging activity by using DPPH, and for their toxicity using brine shrimp sethality Test (BSLT) method. The result of the antioxiant assay showed that n-hexane extract, ethyl acetate extract, CH-5, and CH-6 exhibited 669.48 µg/mL, 12.25 µg/mL, 6.27 µg/mL, 51.87 µg/mL and 7.29 µg/mL, respectively. The toxicity of extracts and the pure compounds recorded as LC50 > 1000 ppm. Therefore, red miana (C.hybridus) is considered as a source of antioxidant agent.
