Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Dari Daun Mempening (Lithocarpus Bancanus)
Mempening (Lithocarpus bancanus) is a plant from Fagaceae family found in the Talang Mamak tribe in Kelayang District, Indragiri Hulu Regency, Riau Province. This plant is usually used by the tribe as a medicine for several diseases such as pain relief and inflammation. Some species of this genus have been reported to contain various secondary metabolites, including terpenoids, steroids and flavonoids. Likewise, bioactivities from the genus have been evaluated including antioxidants, antidiabetic, anticancer antimicrobials and other activities. No such report found from literature regarding this plant. Therefore, metabolites of this plant and their activities were investigated. Isolation the metabolites was started by extraction of the leaves was done by maceration using n-hexane followed by ethyl acetate. The extracts were separated by using gradient solvent VLC and produced 11 fractions from the n-hexane and 10 fractions from the ethyl acetate extract. Brownish-yellow crystals were isolated from fraction 4 of n-hexane (LB-H-03) with the melting point of 175-177oC. White crystals were obtained from fraction 3 of ethyl acetate (LB-E-01) with the melting point of 213-215oC. Compound from LB-E-04, light green crystals was yielded from fraction 176-205 (eluent : n-hexane : ethyl acetate (6 : 4)) with the melting point of 195- 197oC. Furthermore, the extracts and the compounds were determined for their toxicity level by using the Brine Shrimp Lethality Test (BSLT). The results showed that LB-H-03, LB-E-01, and LB-E-04 were toxic with LC50 values of 76.66 ppm, 0.08 ppm, and 25.46 ppm, respectively.
