Isolasi Dan Uji Aktivitas Antimikroba Senyawa Metabolit Sekunder Dari Daun Mahang (Macaranga Bancana)
Macaranga bancana (Euphorbiaceae) is known as "mahang" which is widely distributed in Indragiri Hulu, Riau Province and it is also believed to posses medicinal properties, such as anti-infection agents. This study was to isolate and characterize the secondary metabolites, and their antimicrobial activity from n-hexane and ethyl acetate extracts as well as their fractions. Extraction techniques were applied by using gradual maceration method with n-hexane and ethyl acetate solvents, respectively. Secondary metabolites compounds were separated by using liquid chromatography and the pure compounds were characterized by FTIR and NMR spectroscopies. Antimicrobial activity analyses were done by using microdilution with resazurin as an indicator of microbial growth. White crystal were isolated from n-hexane extract (MB-H-02) with melting point of 224-226oC, while white amorphous solid (MB-E-02) was isolated from ethyl acetate extracts with melting point of 250-251oC. Based on the spectroscopies data and compared to literatures, the compounds were identified as -amyrin palmitate (MB-H-02) and daucosterol (MB-E-02). Antimicrobial analysis showed that n-hexane and ethyl acetate extracts possessed strong activity against S. aureus and C. albicans with 100% inhibitory, while E. coli with inhibitory percentage of 45% and 72%, respectively. Therefore, it could be concluded that mahang possesses good antimicrobial activity and could be used as an anti-infection agent.
