Isolasi Dan Uji Aktivitas Antidiabetes Metabolit Sekunder Dari Akar Tumbuhan Tunjuk Langit (Helminthostachys Zeylanica)
Diabetes is a disease which characterized by hyperglycemia caused by abnormalities of insulin. To treat this disease is to use natural product such as plant. Medicinal plants have long been used by Indonesian people, especially by the Talang Mamak tribe who live around Indragiri Hulu Regency, Riau Province. One of the plants used as traditional medicine is tunjuk langit. This study was to isolate and characterize the secondary metabolites, and their antidiabetic activity from the roots of this species. Extraction techniques were applied by using gradual maceration method with n-hexane and ethyl acetate, respectively. Secondary metabolites were separated by using liquid chromatography and the pure compounds were characterized by FTIR and NMR spectroscopies. Antidiabetic activity tests were carried out using acarbose as a positive control. The compounds were isolated from ethyl acetate extract were a yellow crystal (HZ.K-E-02) with a melting point of 220-222 oC and (HZ.K-E-03) with a melting point of 229-231 oC. Based on spectroscopic data, the compounds were identified as ugonin K (HZ.K-E-02) and ugonin J (HZ.K-E-03). Antidiabetic activity results showed that the compounds had IC50 values 138,6±0,2 µg/mL and 273,3±0,4 µg/mL where the activity value of both compounds was lower than the IC50 of acarbose 0,97±0,02 µg/mL.
