isolasi dan uji aktivitas antidiabetes metabolit sekunder dari kulit batang tumbuhan jelutung (dyera costulata)
Jelutung (Dyera costulata) is a plant of the Apocynaceae family which is found in the Talang Mamak tribal, Kelayang District, Indragiri Hulu Regency, Riau Province. It is usually used by the Talang Mamak tribe as a medicinal plant to treat various diseases such as skin diseases, digestive infections, and throat infections (coughing). The plant contains secondary metabolites such as alkaloids, flavonoids and terpenoids. The species has been reported to have analgesic, antioxidants, anti-osteoporosis, and anti-inflammatory activities. Isolation process of metabolites from the stem bark of this plant was using dicloromethane and n-hexane. The extracts were separated using VLC with a gradient solvent and each extract produced 11 fractions. In fraction 1 of dichloromethane extract, it was obtained a pure compound (DC-D-01) in the form of brownish yellow crystals with a melting point of 124-126 oC assigned as benzoic acid. Fraction 2 of n-hexane extract, it was obtained pure compound (DC-H-01) in the form of white crystals with a melting point of 168-170 oC assigned as lupeol. In this study, the antidiabetic activity analysis was carried out by an enzyme α-glucosidase inhibition method with the p-nitrophenyl-α-D-glucopyranoside (p-NPG) substrate on the pure compounds. The results showed that the DC-D-01 possessed moderate activity with IC50 143,6 µg/mL, whereas the DC-H-01 had strong activity with IC50 43,0 µg/mL.
