Analisis Sekuens Dna Penyandi 26s Rrna Pada Tanaman Cocor Bebek (Kalanchoe X Laetivirens)
ABSTRACT
The gene encoding 26S rRNA (26S ribosomal Ribonucleic acid) is part of a large
sub-unit of structural ribosomal RNA from eukaryotic ribosomes and is included
in the housekeeping gene group that can be used as an internal control in gene
expression tests in plants. The aim of this study was to analyze the DNA
sequences of 26S rRNA encoding in cocor bebek (Kalanchoe x laetivirens) from
Pekanbaru, Riau Province. The research methods included total DNA isolation,
electrophoresis, polymerase chain reaction (PCR), sequencing, and data analysis
using the BioEdit version 7.0, BLASTn, MEGA version 6.0, and Primer3
programs. The DNA sequence encoding 26S rRNA was obtained with a size of
898 bp and has been registered at GenBank with registration number MW286359.
The DNA sequence of the 26S rRNA K. x laetivirens was 98.89% similar and
formed the same branch as K. daigremontiana. The sequence has the potential to
act as an internal control in testing gene expression in duck cocor after validation.
Primers of 26S rRNA for K. x laetivirens have been designed successfully,
namely forward 5'- ATA AGC GGA AGA GAA AC -3 'and reverse 5'- CAG
GCA TAG TTC ACC ATC TT -3' with amplicons measuring 792 bp. These
primers can be used for validation and expression studies.
