Bioprospeksi Bakteri Lokal Pendegradasi Plastik LDPE Dan Oxium

Plastic is a compound that is resistant to degradation and takes a very long time to
completely decompose. The diversity of microbial metabolism allows microbes to
be able to degrade plastics. This study aimed to isolate and characterize endemic
bacteria that have the potential to degrade LDPE and oxium plastics, analyze the
variation of the best bacterial consortium and determine the optimum conditions,
test the ability of the bacterial consortium in biodegradation of LDPE and oxium
plastics at the laboratory and field scales, molecular identification of consortiumforming
bacteria, calculate the effectiveness of environmental costs incurred. from
the application of plastic biodegradation technology. The selection results
obtained 6 isolates of plastic degrading bacteria. Three bacterial isolates that had
the best ability to degrade LDPE and oxium plastics were BP1, BP4, and BP6.
Molecular identification results showed that BP1 had a close phylogenetic
relationship with Sphingobacterium yangliensis, while the bacteria BP4 and BP6
showed a close phylogenetic relationship with Bacillus paramycoides strain 8929.
The best bacterial consortium is consortium 3 (BP1, BP4, and BP6). The optimum
conditions for the biodegradation are pH 7, inoculum concentration of 10%, and
the incubation temperature is room temperature (28-30oC). The percentage of
LDPE plastic degradation by a consortium of bacteria in a laboratory-scale test
was 7.47 ± 0.063% and for oxium plastic 19.09 ± 2.86% within 30 days. The
application of consortium 3 in the form of EM (effective microorganism) on a
field-scale test can accelerate the degradation of LDPE and oxium plastics with
the percentage of degradation of LDPE plastics 7.45±1.26% and oxium plastics
10.17±0.70% within 6 weeks. The results of calculating the efficiency and
effectiveness of environmental costs related to biodegradation technology show
that it is more efficient from an economic perspective than the landfill method. A
review of the ecological, economic, and social aspects related to the application of
LDPE and oxium plastic biodegradation technology shows that these local
bacteria have the potential to be used to overcome the problem of plastic
accumulation in the environment, especially LDPE and oxium.
