Penapisan Bakteri Endofit Asal Bibit Akasia (Acacia Crasicarpa) Dan Uji Antagonisnya Terhadap Xanthomonas Sp. Serta Identifikasinya
ABSTRACT
Acacia (Acaciacrasicarpa) is an industrial plant that has high economic value for cultivation. An important disease in acacia nurseries is bacterial leaf blight caused by Xanthomonassp.Initial symptoms of Xanthomonassp. is a transparent chlorotic spot (slightly yellowish) whose shape is delimited by the bones of the leaves. Controls that have been carried out on Xanthomonas sp. include thinning (spacing), environmental sanitation, biological control using antagonistic microorganisms Pseudomonas fluorescens, Trichoderma sp, and Bacillus subtilis. The aim of this study was to select and test the antagonistic activity of endophytic bacteria from acacia seedlings against xanthomonas sp. in vitro as well as identification of endophytic bacteria based on characteristics, morphology, physiology and biochemistry. Phase 3 research was conducted experimentally using a completely randomized design (CRD) consisting of 11 treatments and 3 replications. The data obtained were analyzed using variance and to compare the mean between treatments, Duncan's multiple distance test was performed at the 5% level. The results showed that 11 isolates of acacia seed endophytic bacteria from different isolates based on the shape of the colony. The results of the hypersensitivity test on 11 isolates of endophytic bacteria on acacia seedlings showed 7 isolates of positive hypersensitivity, namely isolates Ea3, EA4, EA5, Eb2, Eb3, Ed2 and Ed3, but 4 isolates of endophytic bacteria with high antagonistic power were negative hypersensitivity to tobacco plants. Four isolates of endophytic bacteria had high antagonism against Xanthomonas sp. In vitro, isolates Ea1 (root-1) were (5.83 mm), Ea2 (root-2) were (6.33 mm), Eb1 (stem-1) were (7.83 mm) and Ed1( leaf-1) that is (8.50 mm). The results of the characterization of endophytic bacteria of acacia seedlings indicated that isolates Ea1 and Ea2 belonged to the genus Aromonas, isolates Eb1 belonged to the genus Enteribacter and isolates Ed1 belonged to the genus Listeria.
