Efektivitas Penggunaan Larutan Daun Pepaya (Carica Papaya L.) Terhadap Kadar Hormon Kortisol Pada Benih Ikan Nila (Oreochomis Niloticus) Dalam Transportasi
Tilapia is a type of freshwater cultured fish that has good prospects to be developed. To fill the customer’s demand of tilapia, the seed of the fish may be transported from other areas and thus the seed must be transported. During the transportation, however, the seed might be stressed and some of them might be died. To reduce the mortality of the seed during transportation, specific handling technique is needed. The papaya (Carica papaya L.) leaf extract may potential to reduce the stress. Stress response in fish is related to the production of cortisol hormone that is produced by the adrenal glands and regulated by the pituitary gland. By treating the fish using the papaya leaf extract during the transportation, the fish might be less distressed and the cortisol production is reduced.
The aims of this study were to analyze the phytochemical content of papaya leaves, to analyze the effectiveness of papaya leaf extract in reducing the cortisol production and to find out the best dose papaya extract for reducing cortisol during the transportation of tilapia seeds. This study was also aimed to observe the behavior of fish during the transportation.
This research was carried out in May-June 2022. The phytochemical analysis of papaya leaves was carried out in the Integrated Chemistry Laboratory, Faculty of Fisheries and Marine Science, University of Riau. Blood and serum samples were and then were analyzed for cortisol content in the Physiology Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh.
In this study a completely randomized design (CRD) with four treatments and three replications was applied. The treatments applied were as follows: Control with no papaya leaf extract,1.25 ml papaya extract/L water, 2.50 papaya extract/L water and 3.75 ml/L papaya extract/L water. Eight liter of water enriched with papaya leaf extract was put in the plastic bag, filled with 20 fish/bag and sealed. Four bags were tested before being transported and twelve bags were tested after being transported. The tilapia seeds are transported for five hours using cars. Parameters measured were cortisol concentration, survival rate and phytochemical analysis of papaya leaves, namely alkaloids, saponins and flavonoids.
