Induksi Poliploidi Pada Tanaman Acacia Crassicarpa A. Cunn. Ex Benth Dengan Kolkisin Secara In Vitro
Acacia crassicarpa A.Cunn. ex Benth is a type of fast-growing tree species
that is highly adaptable and tolerant of extremely environmental conditions.
Acacia crassicarpa is a type of forest plant that has harvested period of
approximately 5 years. Economically, the acacia is used as a wood source for
processed into wood pulp, paper, and fuel potential for charcoal production. One
of the effort that can be applied to improve genetics quality and wood production
of acacia trees is through induction of polyploidy or chromosomes doubling with
chemical mutation agent such as colchicine. The aims of this study is to induce
polyploidy in vitro of A. crassicarpa shoot tip explants by using colchicine at
various concentrations and soaking time. This research used a factorial
completely randomized design (CRD) with two factors. The first factor is the
concentration of colchicine solution with 5 levels (0%, 0,02%, 0,04%, 0,06% and
0,08%) and the second factor is the immersion time with 3 levels (24, 48, and 72
hours). There were 15 treatment combinations with each treatment repeated 3
times. The data obtained were statistically analyzed by using ANOVA and the
result of variance that significantly influence were continued with Duncan’s
multiple range test at 5% significance level. The results showed that the
combination of colchicine concentration treatment with immersion time had a
significant effect on the percentage of survival explants, number of leaves, number
of shoots, explant height and stomata size of A. crassicarpa explants, but did not
significantly affect on the number of roots, length of root, percentage of rooted
explants and stomata density of A. crassicarpa explants. Colchicine can induce
chromosomal mutations in A. crassicarpa explants cells through in vitro. All
treatment combinations with colchicine were caused polyploidization of
chromosomes in A. crassicarpa cells, but some treatment combination were
showed the same chromosomes number as diploid plant and some less. The result
showed various of chromosomes number, from 24-68 chromosomes per cell, 32
plantlets were obtained which had cells with a higher number of chromosomes
than normal and 3 plantlets that had a lower number. Induction of polyploidy on
A. crassicarpa explants to obtain should be carried out at 0,08% of colchicine
concentration with 72 hours of immersion time.
