Maskulinisasi Ikan Guppy (Poecilia reticulata) menggunakan Ekstrak Daun Karamunting (Rhodomyrtus tomentosa (Aiton) Hassk.)
Poecilia reticulata are popular freshwater ornamental fish and are included in the 6 types of ornamental fish that are in great demand, has high commercial value in domestic and foreign markets. Domestically, the price of guppy fish reaches Rp. 15,000–35,000/head while for foreign countries the price of guppy fish starts from Rp. 275,000–780,000/head. The obstacle in cultivating guppies is that they produce fewer male chicks than female chicks. Therefore, guppy fish cultivation focuses more on the production of male guppy fish. One method that can be applied to increase the production of male guppy fish is the masculinization technique using natural steroid hormones, namely the caramunting plant. Caramunting acts as a steroid or testosterone, stimulating the central nervous system to change gender through sexual differentiation.
This study aims to analyze the levels of the hormone testosterone and estradiol-17β in caramunting leaf extract, analyze the effect and best dose of giving karamunting leaf extract to the parent on the percentage of male sex and the survival rate of guppy fish offspring, analyze the levels of the hormone estradiol-17β in pregnant mothers after soaking in caramunting leaf extract, analyzing the differences in male and female morphology in guppy fish offspring from mothers soaked in caramunting leaf extract.
This research was carried out for 2 months in the Cultivation Engineering Laboratory, Faculty of Fisheries and Marine Sciences, Riau University. The research method used was an experimental method using a Completely Randomized Design (CRD) with one factor, 5 treatments and four replications analyzed in the laboratory and analysis of variance. The dose used is the dose (o, 1, 1.5, 2, 2.5) mg/200ml. Analysis of testosterone and estradiol-17β hormone levels in caramunting leaf extract, analysis of estradiol-17β levels in pregnant guppy fish was carried out at the Laboratory of Physiology, Faculty of Veterinary Medicine, University of Syiah Kuala, Aceh. The parameters measured were analysis of testosterone and estradiol-17β hormone levels in caramunting leaves, analysis of the percentage of male sex and survival rate of guppy fish offspring, analysis of estradiol-17β hormone levels in pregnant guppy fish parents, analysis of differences in the morphology of male and female guppy fish offspring, water quality measurements (pH, DO and ammonia).
The results obtained during the study showed levels of the hormone testosterone and estradiol17-β (ng/g) of caramunting leaf extract in 1 g amounted to 8.34 ng/g and 2.45 ng/g, The highest percentage of males (%) at a dose of 2 mg/200ml water was 65.2 ± 5.9 % and the highest fish survival rate (%) at a dose of 2 mg/200ml water was 100%, the lowest level of estradiol17-β in pregnant mothers (ng/ml-¹) was 0.9 ng/ml-¹ at a dose of 2 mg/200ml. The air quality obtained during the research was that the temperature ranged from 27-28.3°C, the pH value ranged from 6.3-7.2, the DO value ranged from 4.6-6.6 mg/L and the Ammonia value ranged from 0.0197-0.1103 mg/L.
