Produksi Pigmen Penicillium Sp. Lbkurcc27 Pada Media Cair Dengan Variasi Jumlah Inokulum Pada Kondisi Statis Dan Non-Statis
Research conducted by Gaol et al., (2023) had shown that Penicillium sp. LBKURCC27 was able to produce an orange pigment in potato dextrose broth (PDB) media with a pH of 5.5. As one step to increase the level of production of extracellular pigments and intracellular pigments by Penicillium sp. LBKURCC27, this research had two aims. The first aim was to determine the optimal amount of inoculum for Penicillium sp. LBKURCC27 to produce pigment by varying the initial inoculum amount by 1 to 5 plugs in every 50 mL PDB media under static conditions. The second aim was to determine the optimal incubation condition between static and non-static fermentation when pigment production used only 1 inoculum plug per 50 mL PDB media. The results of the research showed that extracellular pigment production under static conditions after 40 days incubation was optimal using 3 plugs of inoculum per 50 mL PDB media, with an average pigment mass of 70.6 ± 0.6 mg/L. Intracellular pigment production under static conditions after 40 days incubation was optimal when using 5 plugs of inoculum per 50 mL PDB media, with a pigment mass of 108.4 ± 8.4 mg/L. Mycelial mass obtained at 3 and 5 plugs per 50 mL PDB media after 40 days incubation was 14.6 ± 0.05 g/L and 15.1 ± 0.1 g/L respectively. Optimum incubation conditions for Penicillium sp. LBKURCC27 to produce pigment in this study was achieved when incubation was carried out non-statically. After 40 days incubation in static condition of Penicillium LBKURCC27 in 50 mL PDB with an initial inoculum of 1 plug, the extracellular and intracellular pigments produced were 49.3 ± 2.3 mg/L and 47.3 ± 4 mg/L respectively, and the mycelial biomass was 11 ± 0.7 g/L.
