Isolasi Dan Uji Aktivitas Antijamur Senyawa Metabolit Sekunder Fraksi Tiga Ekstrak N-Heksana Tumbuhan Paku Tanduk Rusa (Platycerium Bifurcatum)
Platycerium bifurcatum (Cav.) is an epiphytic plant belonging to the Polypodiaceae family, characterized by hanging and branching leaves that resemble deer antlers. Previous research has reported antifungal activity in the ethyl acetate extract of this plant. Based on the literature, no research has examined the antifungal activity of n-hexane extract from the fern P. bifurcatum. This research aims to isolate secondary metabolite compounds from the n-hexane extract of the fern P. bifurcatum and evaluate the potential of these compounds as an antifungal agent. Samples were extracted using the maceration method with methanol solvent, followed by liquid-liquid extraction with n-hexane, dichloromethane, and ethyl acetate solvents. The n-hexane extract was separated using Vacuum Liquid Chromatography (VLC), then fraction 3 was further separated using flash chromatography, separation was continued for subfraction 6 using flash chromatography, and further separation was carried out for subfraction 3 using flash chromatography. The purity of the compounds obtained was tested using Thin Layer Chromatography (TLC), melting point determination, and High-Performance Liquid Chromatography (HPLC). Compound characterization was done using a UV-visible spectrophotometer, Fourier Transform Infrared (FTIR) spectroscopy, and Nuclear Magnetic Resonance (NMR) spectroscopy. The antifungal activity test was conducted using the microdilution method to determine the Minimum Inhibitory Concentration (MIC) and Minimum Fungicidal Concentration (MFC) values. The results of separation using VLC showed five main fractions. Fraction 3 was further separated using flash chromatography to produce thirteen main subfractions, subfraction 6 produced six main subfractions, and subfraction 3 produced twelve main subfractions.
