Isolasi Dan Identifikasi Metabolit Sekunder Dari Fraksi 5 Ekstrak Metanol Benalu Teh (Loranthus Ferrugineus) Serta Uji Toksisitas Menggunakan Metode Brine Shrimp Lethality Test
Loranthus ferrugineus is a parasitic plant belonging to the Loranthaceae family. This plant has been used to cure various human diseases, such as diabetes, inflammation, and cancer. A number of studies have been carried out showing that plants from the Loranthaceae family have antibacterial, antifungal, antioxidant, and anticancer activity. The aim of this research is to isolate and characterize secondary metabolite compounds and carry out toxicity tests using the BSLT method on compounds from F5 of tea mistletoe. Isolation of secondary metabolites has been done using the maceration method with methanol solvent. The methanol extract obtained was separated using vacuum liquid chromatography (VLC) until the final result was obtained in the form of 12 fractions. The fraction obtained was subjected to a thin layer chromatography (TLC) test, and the target compound was F5 in the form of dark green caramel, weighing 27 mg. The compound was analyzed for its purity using TLC and high- performance liquid chromatography (HPLC). The compound obtained was coded LF-M-05 and characterized using a UV-Vis and FTIR spectrophotometer. The UV-Vis measurement spectrum revealed that compound F5 has a maximum absorption at a wavelength of 280 nm, with an absorbance of 0,28. Meanwhile, the FTIR spectrum revealed the presence of an OH group at a wave number of 3369 cm-1, followed by the presence of stretching vibrations in the aliphatic C-H group at a wave number of 2917 cm-1, the C=C group at a wave number of 1605 cm-1, and the alcohol C-O group at a wave number of 1047 cm-1. The toxicity test results, conducted using the BSLT method, show that compound LF-M-05 has an LC50 value of 15595 ppm. Based on the spectroscopy analysis and BSLT test, it can be concluded that the LF-M-05 might originate from secondary metabolite compounds derived from catechins, and it is non-toxic because a compound can be said to be toxic if it has an LC50 value < 1000 ppm.
