Isolasi Senyawa Metabolit Sekunder Dari Fraksi 5 Ekstrak Kloroform Tumbuhan Paku Kaki Tupai (Davallia Denticulata) Dan Uji Aktivitas Antijamur
Davallia denticulata, a member of the Davalliaceae genus, is an epiphytic plant that flourishes on host plants. Several species in this genus have been documented to contain antioxidant, antibacterial, and anticancer characteristic. The objective of this study was to isolate secondary metabolites from the chloroform extract of D. denticulata and evaluate the antifungal properties of the extract, fractions, and isolated compounds obtained. The chloroform extract was prepared by using maceration followed by Vacuum Liquid Chromatography (VLC), resulting in a total of 16 fractions. By combining the fractions derived from Thin Layer Chromatography (TLC) that exhibited identical spots, a total of 7 primary fractions were obtained. Crystals were acquired from subfraction 5 and underwent recrystallization. The recrystallization results on the thin-layer chromatography (TLC) indicated the presence of only one spot on the TLC plate. Additionally, the compound’s purity was assessed using melting point measurement, which revealed a range of 170-172 °C. furthermore, high-performance liquid chromatography (HPLC) analysis demonstrated a single peak, confirming the compound's purity. The resulting compound was assigned the code 23-01-B-FV, and its structural identification was analyzed using UV-VIS, FTIR, and NMR techniques. According to the FTIR and NMR data, the chemical showed similarity to myricetin. The extract, fractions, and compound were subjected to antifungal activity testing using the microdilution method, employing three fungus strains: C. albicans, C. glabrata, and C. krusei. The results obtained showed the smallest MIC value for all extracts, namely 250 ppm and the smallest MFC value for n-hexane, chloroform, ethyl acetate extracts, namely 1000 ppm, while the water extract had the smallest MIC value, namely 500 ppm. The fractions that produced the smallest MIC values were in F5 and F7 with a value of 125 ppm, while the largest MIC values were in F1, F4 and F6 with a value of 500 ppm and the MFC value in all fractions had the smallest value, namely 500 ppm. The compound has the smallest MIC value, namely 25 ppm and MFC value, namely 100 ppm.
