Isolasi, Kultivasi, Dan Potensi Chlorella Sebagai Suplemen Kesehatan Untuk Pencegahan Anemia = Isolation, Cultivation, And The Potential Of Chlorella As A Health Supplement For Anemia Prevention
This research was conducted in November 2020 to December 2023. The objectives of the research were (1) identify and characterize Chlorella vulgaris isolated from the waters of the Rokan River Estuary, Bangko District, Rokan Hilir Regency, Riau Province, (2) evaluate the effect of different composition media on growth characteristics, proximate chemical composition, chlorophyll (a, b), total chlorophyll, carotenoids, antioxidants, and minerals of Chlorella vulgaris isolated from the waters of the Rokan Hilir Regency, Riau Province, (3) characterize the quality of Chlorella vulgaris from the best media cultivation results, (4) determine the chlorophyll quality of Chlorella vulgaris, and (5) investigate the effect of administering different doses of Chlorella vulgaris chlorophyll on hemoglobin and blood erythrocyte content in experimental mice. The test parameters used were DNA molecular genetic test with 18s rRNA primer, yield, growth, specific growth rate, cell density, chlorophyll a, chlorophyll b, total chlorophyll carotenoid; proximate (water, ash, fiber, fat, carbohydrates, protein); minerals (K, Ca, Mg, K, Na), antioxidant activity, amino acids, fatty acids, color intensity, specific chlorophyll profile; hemoglobin and erythrocyte levels of experimental mice. The results showed that the characteristics of chlorella isolated from the waters of Muara Rokan, Rokan Hilir Regency have a green color, an oval shape with a diameter of 2-15 um. Based on GenBank data, it shows the species Chlorella vulgaris. The best formulation media for chlorella cultivation is KNO3 1.50 g/L, KH2PO4 1.25 g/L, MgSO4.7H2O 1 g/L, FeSO4.7H2O 0.0498 g/L which affect the growth, proximate, chlorophyll a, b, carotenoids, total chlorophyll, antioxidants, and mineral content of Chlorella vulgaris. The best Chlorella vulgaris contains 17 amino acids, where the highest essential amino acid is leucine, the lowest is histidine and the highest non-essential amino acid is glutamic acid, the lowest is cysteine; 7 fatty acids, where the highest fatty acid is oleic and the lowest is lauric; and the crude extract contains the highest secondary metabolites in saponins and the lowest in flavonoids; Chlorella vulgaris has a specific chlorophyll profile, namely chlorophyll a, chlorophyll b, pheopitin a and 132 hydroxy chlorophyll a with a presence of >90%; a darker green color than commercial chlorophyll. The best dose for curing anemia in experimental mice is 0.7 mL/KgBW affecting hemoglobin levels and erythrocyte counts for 20 days, where on the 15th day the hemoglobin levels have returned to normal.
