Pengaruh human platelet lysate terhadap proliferasi sel punca mesenkimal
Mesenchymal stem cells (MSC) are cells that proliferate, differentiate and
migrate to damaged tissues. Mesenchymal stem cells can be propagated by
culture methods. Mesenchymal stem cells culture requires supplementation in the
form of serum for MSC nutrition in order to survive and maintain its biological
properties, one of which is proliferation. The serum commonly used in research is
fetal bovine serum (FBS). Human platelet lysate (HPL) is a serum that can be an
alternative to FBS. This study aims to determine the effect of HPL on MSC
proliferation. The design of this study is a laboratory experiment. Mesenchymal
stem cells were isolated from Wharton's jelly, then cultured with HPL and FBS
supplementation for 24 hours and 48 hours. Mesenchymal stem cells proliferation
was assessed using the Cell Counting Kit-8 (CCK-8) method. The difference in the
mean proliferation of MSC in the experimental group was tested using the One-
Way Anova test. To analyze the difference in the mean proliferation of MSC based
on exposure time, a Two-Way Anova test was carried out. The results showed no
significant difference in MSC cultured using 5% HPL, 10% HPL, and 10% FBS
supplementation for 24 hours and 48 hours. Likewise, there was no significant
difference in the MSC proliferation rate based on the incubation time for 24
hours(p value = 0,138) and 48 hours (p value = 0,053). It was concluded that
HPL can be an alternative to FBS as a supplement to MSC culture.
