Analisis Sekuens Nucleotide Binding Site-Leucine Rich Repeat (Nbs-Lrr) Pada Pisang Susu Merah, Pisang Nangka Dan Pisang Batu
Fusarium wilt disease caused by the fungus Fusarium oxysporum f. sp. cubense Tropical Race 4 (Foc TR4) is one of the main diseases of banana plants that can significantly reduce productivity and quality bananas. Previous study showed that three banana cultivars (susu merah, nangka and batu) were potentially resistant to Foc TR4. Therefore, purpose of this study was to analyze NBS-LRR sequence of the three banana cultivars. NBS-LRR has been known related to resistance traits against Fusarium wilt disease. Young banana leaves from the three cultivars were used to isolate genomic DNA following procedures from the Genomic DNA Mini Kit Plant protocol (Geneaid). Electrophoresis result of three samples showed amplicon size of 500 bp using NBSF and NBSR primers. PCR products were sequenced and subsequently converted into deduced amino acid sequences. Conserved Domain Database (CDD) analysis showed that batu banana contains NB-ARC conserved domain and P-loop motif. These result have the potential as genetic markers of resistance in bananas.
