Efek Asam Alfa Lipoat Terhadap Kolagen Matriks Ekstraseluler Sel Β Pankreas Pada Tikus Diabetes Melitus Tipe 2
Many studies have been found using alpha-lipoic acid (ALA) in the preventing of diabetes related chronic complications such as retinopathy, nephropathy, neuropathy, and cardiomyopathy. Langerhans island abnormality in type 2 diabetic (isletopathy) is one of the complications of type 2 diabetes. The extracellular matrix component is an important component for homeostasis, especially the communication of the islet cells and exocrine pancreas cells. The study on the pancreatic extracellular matrix in type 2 diabetic is still limited. This study aimed to analyze the effect of ALA on the collagen of pancreatic β cell extracellular matrix in type 2 diabetic rats. This study was an experimental study with post test only control group design. We used 15 male Rattus norvegicus Wistar strains which were divided into three groups, namely the control group, type 2 diabetic group, and type 2 diabetic+ALA group. Induction of type 2 diabetic was performed by streptozotocin (50 mg/kg body weight) followed by intraperitoneal injection of nicotinamide (110 mg/kg body weight). Alpha lipoic acid was given orally at a dose of 60 mg/kg body weight/day for 3 weeks. Collagen thickness of the pancreatic β cell extracellular matrix was assessed by Verhoeff's van Gieson (VvG) staining with a scoring system. This study showed that in all groups a score of 0 (normal) was obtained. Collagen thickness of the extracellular matrix of pancreatic Langerhans islands remained normal in all study groups.
