Pemurnian Dan Identifikasi Molekuler Fungi Galur J5 Isolat Rizosfer Tanaman Kelapa Sawit Desa Rimbo Panjang, Kampar, Riau Menggunakan Sekuens Its rDNA
Fungal J5 strain was isolated from palm oil’s rhizosphere at Rimbo Panjang Village, Kampar, Riau by Sari (2018). This strain was further purified by the serial dilution method, resulting into two different fungal strains having distinc morphological differences, which were coded as strains LBKURCC310 and LBKURCC311. Morphological characteristics indicated that the fungal LBKURCC310 strain was a member of the genus Trichoderma and LBKURCC311 strain was a member of the genus Polycephalomyces. Morphological identification cannot be used to determine species of fungi with certainty. Molecular identification is required to correctly determine the fungal spesies. Determination of species is very important for studying about ability of each species of fungi to produce enzymes and secondary metabolites. Molecular identification of strains LBKURCC310 and LBKURCC311 were initiated by isolation of genomic DNA from 1 day old mycelia, PCR amplification of ITS-1 and ITS-2 rDNA area sequences using ITS4 and ITS5 primers at annealing temperatures of 42 ºC. The molecular weight of genomic DNA for each isolate were 16,939 and 17,906 bp respectively. While the molecular weight of PCR products for each isolate was 573 and 584 bp respectively. Sequencing of PCR products were done using the Sanger method at the Yayasan Genneka Eijkman Institute for Molecular Biology Jakarta using primers ITS5 (forward), ITS4 (reverse), ITS3 (internal forward) and ITS2 (internal reverse). Sequences were aligned with the Bioedit program to correct gaps or nucleotides whose identity is unknown and analyzed using Mega BLAST, CLUSTAL X v.2.1 and TrichOkey v.2 barcode program for Trichoderma strains. Phylogenetic trees were visualized using the Treeview X and Megaview6 programs. From the Mega BLAST and phylogenetic analysis it was deduced that strain LBKURCC310 was Trichoderma asperelloides and strain LBKURCC311 was Polycephalomyces sinensis
