Perbanyakan Tunas Jeruk Kasturi (Citrus Microcarpa Bunge.) Dari Eksplan Biji Dan Kotiledon Secara In Vitro Dengan Penambahan Bap Dan Gandasil D
ABSTRACT
Kasturi orange (Citrus microcarpa Bunge.) is one of oranges fruit that widely used in daily life. Its demand increased continuously to, but the availability of seeds is limited. Therefore it is necessary to provide seedling in large quantities, one way to propagate the Kasturi plants by in vitro. The purpose of this study was to determine the effect and the best concentration of benzyl amino purine (BAP) and Gandasil D addition using in vitro to the multiplication of shoots from explants of Kasturi orange seed and cotyledon. This research was conducted at the Integrated Laboratory, Department of Biology, Riau University from March to May 2019. The experimental study used explants seed and cotyledon with two separate experiments. This study used a Completely Randomized Design (CRD) with plant growth regulators (PGRs) BAP and Gandasil D. The results showed the addition of BAP and Gandasil D into MS media for the multiplication of Kasturi orange shoots significantly affected the average height and number of leaves in of seed explants, gave no significant effect on shoot multiplication on cotyledonary explants. Concentration of 1 mg/L BAP + 2 mg/L Gandasil D gave the best response with the average time of emergence of shoots (8.50 day after planting), the highest number of shoots (6.75 shoots), the highest shoots (5.92 cm) and the number of leaves (28.50 strands) in seed explants. In cotyledon explants 1 mg/L BAP + 2 mg/L Gandasil D (P7) gave the best response with an average number of shoots (3.00 shoots) and number of leaves (6.00 strands).
