Isolasi Gen Housekeeping Aktin Dan I8s Rrna Pada Durik-Durik (Syzygium Sp.) Dari Danau Kajuik Kabupaten Pelalawan Provinsi Riau

Durik-durik (Syzygium sp.) is one of plants belongs to the family
Myrtaceae that found in Kajuik Lake, Langgam, Pelalawan District, Riau
Province. This plant is well adapted to waterlogging conditions and potential as a
source of tolerant genes of flooding. Gene expression analysis requires internal
controls such as actin and 18S rRNA. This study aimed to isolate actin and 18S
rRNA genes of Durik-durik (Syzygium sp.). Total DNA extraction used Genomic
DNA Mini Kit Plant (Geneaid). Actin gene was amplified using actin primer
specific for Durik-durik and universal primer for 18S rRNA. Data were analyzed
by BLASTn, MEGA Version 7.0, and BioEdit Version 7.0. This study have been
obtained DNA sequences of actin gene with 679 bp in size and it encoded 188
amino acids deduction. The sequences of 18S rRNA gene obtained was 421 bp.
Actin gene sequences of Durik-durik was similar to actin sequences of Gossypium
(ident = 82%). 18S rRNA gene sequences of Durik-durik was similiar to the 18S
rRNA gene sequences of other Myrtaceae family members (ident = 99%).
