Analisis Filogenetik Nibung (Oncosperma Tigillarium (Jack) Ridl.) Asal Kabupaten Bengkalis Berdasarkan Penanda Trnl-F Intergenic Spacer
Nibung (Oncosperma tigillarium) is widely used by coastal communities in Bengkalis Regency. Utilization without conservation efforts to reduce the amount in nature. Taxonomic studies based on morphological characteristics in nibung are difficult because nibung is a plant that takes a long time to grow. So that molecular data support is needed to facilitate taxonomic study. This study aimed to analyze the relationship phylogenetic two collections of nibung and construct phylogenetic trees based on trnL-F intergenic spacer markers. Extraction DNA using CTAB method with modification. DNA sequencing was done in First Base Laboratories Malaysia through PT. Genetics Science Jakarta, Indonesia. DNA sequences are aligned using the Bioedit software. Phylogenetic analysis using the Maximum Parsimony (MP) and Neighbor Joining (NJ) methods using PAUP *. This grouping shows that O. tigiilarium from Bengkalis Island and O. tigiilarium from Rupat Island have a close relationship based on intergenic spacer cpDNA gene. The close kinship between the two nibung collections is due to the geographical location of Bengkalis Island and the nearby Rupat Island. The NJ analysis revealed that O. tigillarium from Rupat Island is more primitive than O. tigillarium from Bengkalis Island due the longer genetic distance. Both Parsimony and Neighhbour Joining supported the close relationship between two collection O. tigillarium from Bengkalis regency.
