Uji Patogenesitas Bakteri Edwardsiella Tarda Pada Ikan Jambal Siam (Pangasionodon Hypophthalmus)

ABSTRACT
Edwardsiella tarda is a pathogenic bacteria that causes a decrease in the
productivity of striped catfish (Pangasionodon hypophthalmus) cultures. It is
necessary to do a pathogenicity test to determine the level of malignancy of
bacteria E. tarda in infecting striped catfish (P. hypophthalmus). The research was
conducted in September-November 2020 at the Fish Quarantine Center for
Quality Control and Safety of Fishery Products in Pekanbaru. The purpose of this
study was to determine the density of E. tarda bacteria which can kill 50% of the
striped catfish (P. hypophthalmus). The method used was an experimental method
with a completely randomized design (CRD) consisting of 4 levels of treatment
and 3 replications. Striped catfish (P. hypophthalmus) used measuring 8-12 cm,
totaling 10 fish with a container volume of 15 L. Adaptation was carried out for 7
days with commercial feeding of ff-999 (35%) which was given adlibitum 3 times
a day at 08.00, 12.00 and 16.00 WIB. Injection is carried out as much as
0,1 mL/head in the lower part of the stomach with P0 treatment (fish not infected
with E. tarda bacteria), P1 (106 CFU/mL), P2 (107 CFU/mL) and P3 (108
CFU/mL). The results showed that the density of E. tarda bacteria which could
kill 50% of the striped catfish (P. hypophthalmus) within 72 hours was 6,7 x 106
CFU/mL. The highest mortality was in P3 (108 CFU/mL), namely 86,88% with
clinical symptoms in the body including ulcers/sores, pale eyes, wrinkled fins, and
pale stomach, intestines, kidneys and liver. The lowest Post-infection erythrocyte
total was at P3 (108 CFU/mL) which was 1,46 x 106 cells/mm3 and the highest
leukocytes total was at P3 (108 CFU/mL) which was 10,94 x 104 cells/mm3. The
quality of water during the study was a temperature of 28,5-29oC, pH 6, DO 6,3-
5,5 mg/L and ammonia 0,05-0,08 mg/L.
