Isolasi Dan Uji Toksisitas Ekstrak N-Heksana Dari Daun Bonai Antidesma Bunius

Antidesma bunius is a plant species belonging to the family Euphorbiaceae. This plant is widely distributed across various tropical regions, including Asia, Africa, the Americas, and the Pacific Islands, with approximately 150 species reported. This study aimed to isolate and characterize a pure compound obtained from the n-hexane extract of A. bunius leaves and to evaluate its toxicity using the brine shrimp lethality test (BSLT) method. The isolation of secondary metabolites from this plant was carried out through flash column chromatography. A total of 134 vials were collected from the flash chromatography process; vials numbered 11–27 were combined into a single fraction due to identical Rf values and subsequently subjected to further separation using radial chromatography (Chromatotron®). The radial chromatography yielded 80 vials, of which vials 19–31 exhibited identical spot patterns and were therefore pooled and recrystallized to obtain a purified compound. This compound was designated AB-HX-K1-19-31, and its purity was evaluated by thin-layer chromatography (TLC) and melting point determination using a Fisher John apparatus. The results indicated that the compound was not yet completely pure, as evidenced by the presence of two spots on the TLC chromatogram and a melting point range of 112–115°C. UV-Vis spectrophotometric analysis revealed a maximum absorption at a wavelength (λmax) of 205 nm with an absorbance of 0.891. FTIR spectroscopic analysis showed characteristic absorption bands at 3308 cm⁻¹ (O–H), 2927 and 2852 cm⁻¹ (aliphatic C–H), 1716 cm⁻¹ (C=O), 1458 and 1375 cm⁻¹ (C–H), 1651 cm⁻¹ (C=C), 1094 cm⁻¹ (C–O), and 2094 cm⁻¹ (C≡C). Furthermore, GC-MS analysis revealed three peaks, which were tentatively identified as ageratriol, patchouli alcohol, and trans-13-octadecenoic acid. The toxicity assay using the BSLT method indicated that the extract was moderately toxic to Artemia salina larvae, with an LC₅₀ value of 343.8 ppm.
