EFFECT OF HYPOXIC PRECONDITIONS ON MESENCHYMAL STEM CELL PROLIFERATION LEVEL AND VIABILITY

Mesenchymal stem cells (MSCs) are potential to be developed as therapeutic agent due to their self-renewal, differentiation, and biological factors secretion capacity. A culture method is important to obtain sufficient amount of cells for therapy. Culture using normal oxygen concentration may effect cell proliferation and viability. Therefore, this study aimed to determine the effect of hypoxic preconditions on the levels of proliferative and viability of MSCs. The research design was a quasi-laboratory experiment with a post-test control group design. The umbilical cord was obtained from caesarean section women, aged 20-35 years and not infected with hepatitis B or HIV. MSCs was isolated from Wharton's jelly of umbilical cord using the explant method. Hypoxia was induced using cobalt chloride (CoCl2) for 24 hours. MSCs proliferative activity was evaluated by the MTT assay method and cell viability was evaluated by the trypan blue exclusion test method. The differences on the rate of proliferation and viability of MSCs cultured under hypoxia and normoxia condition were analyzed by the independent t-test. The results showed a decrease in the rate of SPM proliferation in the hypoxic group compared to the normoxic group (p = 0.001). However, cell viability between the two groups did not show a significant difference (p = 0,425). In conclusion, hypoxia decreased the proliferation rate, but did not affect cell viability of MSCs.
