Efek Suplementasi Vitamin D Terhadap Proliferasi Sel Punca Mesenkimal
Mesenchymal stem cells (SPM) are multipotent progenitor cells with the ability to self-renewal and differentiate into various cells, making them potential for degenerative disease therapy. The SPM culture method is important to multiply the cells so that they are sufficient to obtain therapeutic effects. One of the challenges in SPM culture is to maintain SPM proliferation during the culture process for a long period of time. This study aims to determine the effect of vitamin D supplementation on SPM proliferation. The design of this study was quasi- experimental laboratory. SPM were isolated from umbilical cord wharton's jelly using the explant method, then cultured with or without vitamin D supplementation for 24 and 48 hours. SPM proliferation was evaluated by CCK-8 assay. Differences in mean SPM proliferation based on exposure time between treatment groups were tested using one-way Anova test and Tukey's multiple comparisons test. To analyse the interaction between vitamin D concentration and exposure time on SPM proliferation, two-way Anova test and Sidak's multiple comparisons test were conducted. Determination of EC 50 vitamin D using GraphPad Prism 9.0. The results showed that vitamin D supplementation with concentrations of 0.1; 1; 10 nM for 24 and 48 hours consistently decreased SPM proliferation (p
