Isolasi Senyawa Aktif Antibakteri Dari Ekstrak Etil Asetat Daun Lampeni (Ardisia Humilis Vahl)
Infections caused by microorganisms such as bacteria, are becoming increasingly difficult to treat due to improper use of antibiotics leading to bacterial resistance. Therefore, new antibacterial agents are needed, and one continuously developed source is plants. One potential herbal plant is lampeni (Ardisia humilis Vahl) from the Primulaceae family and Ardisia genus, with several species reported to have antimicrobial activity. This study aims to isolate antibacterial active compounds from the ethyl acetate extract of lampeni leaves and test their activity. The simplisia was macerated with ethyl acetate solvent, followed by fractionation using VLC and resulting in 14 fractions. Fraction F5, which exhibited the best antibacterial activity, was further separated using flash chromatography and yielding 17 subfractions. The combined subfractions 7-11 (FC7-FC11) showed the best antibacterial activity and were isolated using preparative HPLC, resulting in an isolate compound L-B as a pinkish-white solid (14.23 mg). The purity analysis of the compound using analytical UPLC revealed a dominant peak at a retention time of 17.82 minutes. The isolate compound L-B was identified through LC-HRMS, 1H-NMR, and 13C-NMR as a fatty acid derivative (m/z 378.27), consisting of 38 hydrogen atoms, 23 carbon atoms, and 4 oxygen atoms.This identification was supported by FTIR spectrum analysis showing absorptions at wave numbers characteristic of unsaturated fatty acid compounds: 3424 cm-1 and 3315 cm-1 (O-H); 2915 cm-1 and 2848 cm-1 (C-H); 1732 cm-1 (C=O); 1629 cm-1 (C=C); and 1469 cm-1 (CH3). The extract was categorized as bactericidal if the MBC to MIC ratio was less than or equal to 4. Antibacterial activity tests using the microdilution method showed that isolate compound L-B had an MBC/MIC ratio of (2:1) against S. aureus and an MBC/MIC ratio of (1:1) against P. aeruginosa, thus categorizing isolate compound L-B as bactericidal.
