Analisis Metabolomik Dan Kemometrik Ekstrak Fermentasi Ko-Kultur Dari Jamur Penicillium Sp. Lbkurcc34 Dan Lbkurcc39
Infections caused by pathogenic bacteria are still a public health problem in the world. Therefore, the search for sources of bioactive compounds, especially antimicrobials and antioxidants, continues along with the increasing resistance of bacteria to existing antibiotics. One of the fungi isolated from local peat soil in Riau is Penicillium sp. LBKURCC34 and LBKURCC39 which has antimicrobial and antioxidant activities. However, the compounds contained and their role in these activities are not yet known. This study aims to determine the compounds from acetone, ethyl acetate and butanol extracts of Penicillium sp. LBKURCC34 and LBKURCC39 co-cultures that can be utilized as antimicrobials and antioxidants with target and nontarget approaches, which are carried out using Xcalibur, MS-DIAL, GNPS and XLSTAT software to see the correlation between extracts and their activity tests. Analysis with Xcalibur software obtained dominant peaks. The results obtained from Xcalibur analysis in the form of chromatograms show that acetone extracts and butanol extracts dominantly have polar compounds, while ethyl acetate extracts have more peaks distributed from polar, semi-polar and non-polar compounds. Based on the analysis conducted on MS-DIAL, there are 1641 compounds before filtering and filtered compounds that have been annotated as many as 147 compounds. The compound data identified in Xcalibur, MS-DIAL and GNPS are Tryptophan compounds and also several other common natural compounds. Correlation analysis of identified compounds with their activity data using XLSTAT from several selected common natural compounds that correlate strongly with antimicrobial activity, namely 4-Aminophenol, 3-(propan-2-yl)-octahydropyrrolo[1,2-a)pyrazine-1,4-dione, Pentobarbital, NP-011220, Cyclo(phenylalanyl-prolyl), Diftalone, Quwenzhi, 4'-Methoxy-3-morpholino-propiophenone, SCHEMBL23157347, (-)-nabilone, 4-{(Z)-[(4E, 7Z, 16Z, 19Z)-1-Hydroxy-4,7,10,13,16,19-docosahexaen-1-ylidene]amino} butanoic acid and Oleamide. The compounds that have antioxidant activity are Betaine and Panthenol.
