Analisis Metabolomik Dan Kemometrik Ekstrak Fermentasi Ko-Kultur Dari Jamur Penicillium Sp. Lbkurcc34 Dan Lbkurcc38
Fungi are one of the promising microorganisms for the discovery of new drug candidates as antimicrobial and antioxidant compounds. One of the microorganism that is thought to be used as a producer of new antimicrobials is Penicillium sp. LBKURCC34 and LBKURCC38 from the peat soil of the primary forest of the Giam Siak Kecil Bukit Batu Biosphere Reserve (GSKBB), Riau Province. The metabolomic analysis approach is carried out to identify secondary metabolites and see the correlation of compound content with its unknown bioactivity and the limited yield of extracts obtained from microbial fermentation is also a challenge in conducting analysis. Therefore, this study aims to identify compounds from Acetone Extract, Ethyl acetate Extract and Butanol Extract from the fermentation of Penicillium sp. LBKURCC 34 and LBKURCC 38 fungal co-cultures with metabolomic and chemometric analysis approaches using Xcalibur, MS-DIAL, Massive GNPS and XLStat software. Analysis using Xcalibur software obtained chromatograms of acetone and butanol extracts containing polar dominant compounds because they were identified at the initial retention time (1-2 minutes) and chromatograms of ethyl acetate extracts containing compounds containing polar and semi-polar dominant compounds distributed at the initial and middle retention times (1-16 minutes). Analysis using MS-DIAL software identified 3 compounds from the sample extract namely Cyclo(Val-Pro), Cyclo(Tyr-Ala) and Sulindac. Phytosphingosine and Cyclo (Val-Pro) compounds were identified repeatedly in the analysis using Xcalibur, MS-DIAL and GNPS software. Y-Aminobutyric acid (GABA), N-Acetyltyramine, Zindrotine, Supinine, Esmolol, Glycochenodeoxycholic acid and Cyclo(Val-Pro) were strongly correlated as anti S. aureus, anti S. epidermidis, anti E. coli and anti C. albicans. Compounds
L(-)Methionine, N-(2-Phenylethyl)-beta-D-glucopyranuronosylamine and L-Isoleucine correlated quite strongly as anti S. epidermidis.
