Ekstraksi zat pengatur tumbuh alami dari bonggol pisang dan uji aktivitasnya terhadap pertumbuhan pisang kepok (mUSA ACUMINATA X mUSA BALBISIANA) secara in vitro
The addition of synthetic PGRs, especially in in vitro media, has been widely reported with various advantages and disadvantages. Although synthetic PGRs are more practical in their application and their PGR content is known for certain, synthetic PGRs have several disadvantages such as being quite expensive and also having side effects on health and the environment caused by residues from the use of these chemicals. In line with this development, efforts to find natural PGR sources must continue to be carried out to replace synthetic PGRs used in in vitro propagation media. In line with this development, efforts are needed to find natural PGR sources from banana stems and test their effectiveness on kapok bananas (Musa acuminata x Musa balbisiana) in vitro. This study consists of the stages of extracting natural PGRs from kepok bananas and testing their activity on kepok banana explants in vitro.
The research was conducted at the Natural Material Organic Chemistry Laboratory, Department of Chemistry, Faculty of Mathematics and Natural Sciences, University of Riau, and the Tissue Culture Laboratory, Department of Agriculture, Riau Province. The study used ZPT extraction with the KCKT technique and an In Vitro research design using a Completely Randomized Design (CRD) consisting of different doses of ZPT extract. I0: 0 mg.l-1, I1: 0,2 mg.l-1, I2: 0,6 mg.l-1, I3: 1,8 mg.l-1, I4: 5,4 mg.l-1. There were 5 treatments and each treatment was repeated 5 (five) times to produce 25 experimental units (bottles). Each experimental unit (bottle) consisted of 1 (one) banana corm explant. The data obtained were analyzed using ANOVA and then further tested with DMRT at a level of 5%.
The results showed that the methanol extract of banana stump showed positive results for the presence of plant growth regulators, namely gibberellin with a concentration of 3,866 mg.l-1, auxin with a concentration of 2,177 mg.l-1 and cytokinins in the form of kinetin and zeatin with concentrations of 0,877 mg.l-1 and 0,712 mg.l-1 respectively. The administration of natural growth regulator extracts from banana stumps at various concentrations resulted in a percentage of living explants of 100%. The addition of 0,6 mg.l-1 of natural growth regulator extract from banana stumps gave the best results on shoot growth, namely accelerating the time of shoot emergence at 67,00 hst, the number of shoots was 1,40, the length of shoots was 7,56 cm and produced 1,8 leaves. The administration of natural growth regulator extracts from banana stumps at a
concentration of 1,8 mg.l-1 gave the best results on root growth, namely accelerating the time of root emergence at 62,25 hst, with the longest root being 22,25 cm.
